Sakural Japanese Foundation for Cancer Research, Cancer Chemotherapy Center, Toshlma-ku, Tokyo 170, Japan We reported previously that the combination of concanavalin A(ConA)-bound, but not ConA-free, LI210 murlne leukemia vaccine and pyran copolymer produced an enhanced therapeutic response in Ll210-bearlng mice. Immunoprophylactic experiments had shown that their effective combination induced the enhanced antltumor T cell production in mice after, but not before, llve LI210 inoculation. In the present study we characterized Immunocytes associated with the induction of enhanced antitumor immunity and found that amplifier T cells induced by ConA-bound LI210 vaccine was partly responsible for enhanced antitumor immunity as evidenced by the following findings. Amplifier T cells were induced in spleen of mice sensitized with both agents. When these mlcewere splenectomyzed, they were not resistant to live LI210 inoculation. Mice sensitized with ConA-free Ll210 vaccine and pyran copolymer or mice sensitized first with pyran copolymer and subsequently with ConA-bound L1210 vaccine in which amplifier cells were not induced were not resistant to live L1210 inoculation either.
