N2 - The RNAs of different influenza viruses were analyzed together with marker RNAs on polyacrylamide-urea gels after denaturation with glyoxal. Using this new method for determining molecular weights (MW) the following results were obtained: (1) The total MW of the RNA of human and animal influenza A viruses was found to be 4.9 x 106, and the MW of influenza B virus RNA was 5.3 x 106. (2) After glyoxalation migration patterns of the RNAs coding for HA and NA varied among influenza A viruses belonging to different subtypes, while the mobility of corresponding RNAs coding for the Pl, P2, P3, NP, M, and NS proteins was identical. This suggested that the size of the latter genes was conserved among different human and animal influenza A viruses but that the size of the HA and NA genes may vary. Among strains containing Nl neuraminidase three different migration patterns of the neuraminidase gene were observed. (3) After glyoxalation no migration differences were observed among corresponding RNA segments of different influenza B viruses.
