Immunohistochemical evidence of the expression and localization of glucose transporter in brain neurons and glia cells in rats

Abstract

Aim: To explore the distribution of Mr 45 000 glucose transporter 1 in the brain parenchymal cells. Methods: The experiment was finished in the Laboratory of Nerve Biologic Research of Institute of Hypoxia Medical science of Xuanwu Hospital of Capital University of Medical Sciences from August to September 2004. Two two-grade SD adult male rats were selected with 250 g body mass. After the make of homogenate using one rat brain, the specificity of antiserum which was made by glucose transporter 1 C-end part synthesis with peptide immune rabbits on M, 45 000 glucose transporter 1 was tested with immunoblot trial. Another male rat under deep anesthesia, brain was separated and made into 20 μm section. The localization of M, 45 000 glucose transporter 1 in the brain parenchyma cells in rats was observed by antibiotic protein-biotin complex staining and immunoelectron microscopy. Results: 1 The antiserum made by glucose transporter 1 C-end part synthesis with peptide immune rabbits was a high effectively unique antibody on M, 45 000 glucose transporter 1. 2 The M, 45 000 glucose transporter 1 protein expressed in some particular neurons of the normal brain glial cell and except cortex. 3 Most of positive glial cells of M, 45 000 glucose transporter 1 were oligodendrocytes, and a little of them were astrocytes. Conclusion: Based on the elongation of the reaction time of antibody and brain tissue section, the Mr 45 000 glucose transporter 1 expressed in astrocyte, oligodendrocyte and some particular parts of neurons.
