N1 - Funding Information: If the antisense probe is appropriately radiolabeled, it should allow a noninvasive diagnostic strategy for evaluation of mRNA associated with the disease process. For the development of effective imaging strategy, it is necessary that there is sufficient increase in mRNA production, that the antisense oligonucleotide (ASON) probe is appropriately radiolabeled at high specific activity and is able to negotiate through the cell membrane into the cytoplasm. The ASON probe should be stable against degradation from nucleases and should comprise high enough specific activity to detect relatively low levels of From the Departmento f VeterinaryB iosciences,U niversityo f Illinois, Urbana, Illinois, and aAllegheny University Hospital, Hahnemann Division, Philadelphia, Pennsylvania. Regardless of whether the gene of interest is an endogenous gene or exogenously administered (gene therapy), the same type of probes are used for the imaging of gene expression. The radiolabeled probes administered at microgram level may not affect transcription.
