Analysis of 8-hydroxydeoxyguanosine in human urine by gas chromatography/mass spectrometry

Abstract

A method of analysis of 8-hydroxydeoxyguanosine (8OHdG) in human urine by gas chromatography/mass spectrometry was developed. On the basis of the qualitative analysis of MS, the most intense ion at m/z 383 corresponding to the base + 1 (B + l) was the characteristic ion of 8OHdG. When the selected ion mode (GC/MS-SIM) was chosen as the quantitative method, the detection limit of this method was 0.5 nmol/L, and the linear range was 5-10000 nmol/L. The average interday and interday coefficients of the variation of the method were 4.23% and 8.25%, respectively. For analysis of urinary 8OHdG, the recovery of spiked urine is 99.37% and the RSD is 5.12%. Using the developed method, the excretion levels of urinary 8OHdG of two healthy Chinese persons and six patients from three kinds of cancer were determined. It was found that the excretion levels of urinary 8OHdG in cancer patients were significantly higher than that of urinary 8OHdG in healthy persons.
