Rational Design of Effective Prostate Cancer Combination Therapies

Despite the availability of several new targeted therapies for prostate cancer. treatment of patients is ultimately impeded by the development of treatment resistance. Efforts to circumvent resistance typically rely on various drug cocktails that combine available therapeutics. However, the overall impact of anti-cancer drug interactions—including their influence on treatment resistance—remains poorly understood. Defining such interactions is critical for designing the most effective combination therapies. especially as new drugs are approved. Because there is no cure for advanced prostate cancer. the disease warrants the development of aggressive and results-oriented treatment strategies that extend the survival of patients without compromising quality of life. In ordor to address this pressing issue. Dr. Yadav has designed a three-phase research program to identify ideal drug combinations that will prevent or delay the onset of resistance. First, he will analyze interactions among existing prostate cancer drugs in order to identify potential drug-pairs. Using state of the art genomics„ he will then characterize the mechanism(si of drug resistance. Ultimately. he wilt generate a database of drug resistance signatures, focusing on recurring genetic alterations that confer resistance to clinically relevant drugs. Subsequently Or. Yadav will test whether combining drugs with minimal overlap in their drug-resistance signatures can delay the emergence of resistance. Finally, he will establish a small-scale clinical trial to assess efficacy of combination therapy.
